NMR occupies a prominent place in analytical chemistry and structural biology due to its ability to probe matter at the atomic scale. Its main limitation is its low sensitivity. In this thesis, we will propose an original hyperpolarisation solution and evaluate its field of application on a series of molecules ranging from natural products to therapeutic peptides and proteins.
L'objectif principal de la thèse sera de proposer une nouvelle méthode pour augmenter significativement le signal RMN de molécules. La technique photo-CIDNP consiste à illuminer un échantillon contenant un colorant aux propriétés photophysiques adéquates, pour le faire passer dans un état excité de type triplet. Les interactions moléculaires entre le colorant et la molécule à hyperpolariser permettent ensuite le transfert de polarisation vers la molécule d'intérêt, boostant ainsi le signal RMN. Cependant, à ce jour, les techniques photo-CIDNP mènent à l'hyperpolarisation que d'un nombre limité de spins 1H ou 13C sur une molécule, limitant d'autant la capacité de la technique. Durant la thèse, nous viserons à mettre au point plusieurs méthodes complémentaires pour diffuser cette hyperpolarisation le long de la molécule d'intérêt et donc d'augmenter les signaux d'un plus grand nombre de spins/atomes. Nous déclinerons ces méthodes pour améliorer la détection de petites molécules ou peptides dans des mélanges complexes ou pour mieux caractériser la surface des protéines en lien avec des modifications chimiques et post-traductionnelles. En parallèle, un développement de méthode orthogonale sera effectué pour la caractérisation des produits par HPLC ou toutes autres techniques biophysique.

The main objective of the thesis will be to propose a new method for significantly increasing the NMR signal of molecules. The photo-CIDNP technique consists of illuminating a sample containing a dye with suitable photophysical properties, causing it to enter an excited triplet state. The molecular interactions between the dye and the molecule to be hyperpolarised then allow the polarisation to be transferred to the molecule of interest, thereby boosting the NMR signal. However, to date, photo-CIDNP techniques only lead to the hyperpolarisation of a limited number of 1H or 13C spins on a molecule, thus limiting the technique's capacity. During the thesis, we will aim to develop several complementary methods to spread this hyperpolarisation along the molecule of interest and thus increase the signals from a larger number of spins/atoms. We will adapt these methods to improve the detection of small molecules or peptides in complex mixtures or to better characterise the surface of proteins in relation to chemical and post-translational modifications. In parallel, the development of an orthogonal analytical method will be performed to further characterize the products using HPLC and/or other biophysical techniques.
La méthode principale utilisée durant la thèse sera la résonance magnétique nucléaire (RMN). Le/la doctorant·e aura accès à l'ensemble des équipements RMN de la plateforme RMNHC@UPSAY de l'ICSN : 5 spectromètres de 600 à 950 MHz, équipés de cryosonde pour la plupart. L'originalité du projet de thèse résidera dans la maîtrise et l'amélioration de méthodes d'illumination in situ par des LED/laser. Le coeur de la thèse consistera à améliorer le dispositif technique d'illumination, l'optimisation de séquences d'impulsions RMN, ainsi que l'optimisation des échantillons en termes de concentrations et compositions. Les molécules d'intérêt sont pour l'essentiel commerciales. Les protéines enrichies en isotopes stables 15N/13C seront produites au laboratoire, avec ou sans l'intervention du/de la doctorant·e.

The main method used during the thesis will be nuclear magnetic resonance (NMR). The PhD student will have access to all the NMR equipment at the ICSN's RMNHC@UPSAY platform: five spectrometers ranging from 600 to 950 MHz, most of which are equipped with cryoprobes. The originality of the thesis project will lie in the mastery and improvement of in situ illumination methods using LEDs/lasers. The core of the thesis will consist of improving the technical illumination device, optimising NMR pulse sequences, and optimising samples in terms of concentrations and compositions. The molecules of interest are mainly commercial. Proteins enriched with stable isotopes 15N/13C will be produced in the laboratory, with or without the involvement of the PhD student.
